介紹

免疫測定是一種生化測試，用于測量生物體液中蛋白質的濃度。該檢測利用了抗體與其抗原的特異性結合。

有不同類型的免疫測定，該協(xié)議包含有關夾心ELISA（酶聯(lián)免疫吸附測定）的信息。在三明治免疫測定中，也稱為“非競爭性 ELISA"，抗原與抗體位點結合，標記抗體與抗原結合。

測量位點上的抗原量。非競爭性 ELISA 方法的結果將與抗原的濃度成正比。這是因為如果未知樣品中不存在抗原，標記抗體將不會結合。

程序

在測定之前，兩種抗體制劑都應純化且不含白蛋白。檢測抗體必須貼標。

2.通過向每個孔中加入約100μl包被抗體溶液來涂覆孔。使用的抗體量將取決于單個檢測、優(yōu)化緩沖液和包被濃度（1-10 μg/孔）。

4.微量滴定板上蛋白質結合的其余位點必須通過與封閉緩沖液孵育而飽和。用200μl封閉緩沖液填充孔。

5. 用粘性塑料蓋住板，在室溫下孵育 60-90 分鐘，或在 4°C 下孵育過夜。

用洗滌液洗滌孔四次。

7.向孔中加入100μl稀釋的樣品，標準品和對照品。所有稀釋都應在稀釋緩沖液中進行。

8. 用粘性塑料蓋住板，在 37°

C.9 下孵育 60-90 分鐘。用洗滌液清洗盤子四次。

與一抗一起孵育

10. 加入 100 μl 一抗。要添加的量可以在初步實驗中確定。為了準確定量，一抗應過量使用。所有稀釋都應在稀釋緩沖液中進行。

11. 用粘性塑料蓋住板，在室溫或 -20°C 下孵育 60-90 分鐘。

用洗滌液洗滌四次。

與二抗（或生物素化一抗的鏈霉親和素-HRP）孵育

13. 加入 100 μl 標記的二抗。要添加的量可以在初步實驗中確定。為了準確定量，應過量使用標記的二抗。所有稀釋都應在稀釋緩沖液中進行。

14. 用粘性塑料蓋住板，在室溫或 -20°C 下孵育 60-90 分鐘。

15.用洗滌液洗滌四次。

16. 按照制造商的指示添加基材。

17. 在建議的孵育時間過后，向每個孔中加入終止溶液。

18.添加終止液后30分鐘內(nèi)，可在ELISA讀數(shù)儀上測量目標波長的光密度。

數(shù)據(jù)分析

19. 計算每組重復標準品、樣品和對照品的平均吸光度值。

20. 如果單個吸光度值與相應的平均值相差超過15%，則認為結果可疑，應重新測定樣品。

21. 使用能夠生成良好曲線擬合的計算機軟件減少數(shù)據(jù)，從而創(chuàng)建標準曲線。

22. 如果樣品已被稀釋，則根據(jù)標準曲線測定的濃度必須乘以稀釋因子。
